Usage And Features of Water Bath Equipments

We deal with various laboratories equipments and one of them is water bath equipments. It is a device that maintains water at a constant temperature that is used in microbiological laboratory for incubations.

These equipments are provided with several functions that enable you to understand its usage and utility. These functions are more or less the same. Water bath equipments can be of several types. They are double walled water bath, serological water bath, high precision water bath, circulatory water bath, refrigerated water bath and paraffin water bath. These equipments differ only on the basis of their usage.

These equipments have temperature controllers that help to maintain uniformity in the device. Indirect heating is provided in the water bath, which comprises of air heaters made of high grade. The temperature is evenly distributed throughout the chamber through natural water convection mechanism that ensures very good temperature sensitivity. This is a repetitive process.

These equipments only differ on the basis their usage. They are provided with the feature to adjust or select baud rate for any serial communication port. It even incorporates multiple devices such as in line process indicators, environment scanners and temperature or humidity controllers. These equipments are even provided with user friendly custom developed software which obtains and analyses data and facilitates the user to generate report and graphs. This is considered as an ideal module for pharmaceutical laboratories, process control applications and high research projects where maintaining a viable record of the equipment is very important.

Adding on, high precision water bath has a unit fitted with high efficiency stirrer motors of 1/20 HP so as to maintain the uniform temperature of our high precision water bath. Whereas, refrigerated water bath equipment has an energy efficient cooling unit that is installed in our carbon dioxide incubators to enable bio chemical demand studies to enable bio chemical demand studies at lower temperature.

Paraffin section mountain bath has been designed for ease of use and maximum safety. The top has generous space for resting the hands and a deep tapered bowl allowing slides to be used at any angle. The four rubber feet and the lower level of the whole instrument provide stability.

Clean Room and Air Shower

Clean Rooms are designed to restrict the admission of all types of microbial and particulate contamination. A Clean Room is a room that has a controlled level of contamination usually specified by the number of micro particles per cubic meter at a specified size.  Clean rooms are used for a wide variety of industrial, medical, pharmaceutical and research application. Sometimes numerous contaminants and particles can be introduced into a clean room and for that reason clean rooms will feature decontaminating personnel/equipment such as Air Shower. Clean Rooms rely on the effectiveness of terminal high efficiency particulate air HEPA filter (provided by Air Shower) to provide particle free air to the controlled environment. Horizontal Autoclave

Weiber designed horizontal autoclave can be rectangular and cylindrical and is suitable for various applications such as in hospitals, pharmaceuticals, tissue culture and research laboratories. It is double walled unit in which the chambers, boiler, lid and jacket all are made up of SS304 and this whole unit is mounted on duly painted with attractive stoving enamel robust tubular stand made up of mild steel pipes. In this, the heating is provided by ISI mark bearing immersion type heaters and the working pressure and temperature  inside the chamber is 5 psi to 20 psi and 121o C/134o C (depending on the time duration of sterilization) respectively. It  is provided with a steam generator attached underneath jacket, pressure and compound gauge indicates jacket and chamber pressure/vacuum respectively.

Vertical Autoclave

Weiber vertical autoclave; double walled chamber with its outer wall of thick gauge mild steel, inner wall of SS304 grade and lid made up of mild steel with the inner side of unit lined with stainless steel for double protection providing the Radial Locking arrangement for lid. It contains inbuilt safety valves, pressure gauge, pressure releasing valve & a foot paddle lifting device, in this unit the pressure is adjustable from 5 psi to 20 psi and the working temperature is 121o C/134o C, depending on the time period of sterilization. In this, heating is provided through special immersion type heaters.

Portable Autoclave

Weiber portable autoclave made up of Stainless steel is based on the principle of pressure cooker and is suitable for moist heat sterilization of small equipments useful in clinics, hospitals and research and development laboratories. It contains pressure gauge for adjusting pressure inside the chamber, steam releasing valve and is fully equipped with safety valves. It is provided with Bakelite handles to carry the unit and three leg stand.

Extraction and Digestion Unit

Kjeldahl Distillation Unit

Weiber Kjeldahl distillation unit; suitable for the determination of organic nitrogen hence the protein content by the relation % Protein = 6.25 x % using Kjeldahl method that consists of three steps; digestion, distillation and titration. It is made up of knitted layer of glass fabric combined with flexible heating element, encased in Mild Steel housing wrapped by the flexible heater, another layer of glass cloth is placed on the lower mantle held firmly by a circular aluminum ring. It has a minimum of 3 and maximum of 6 mantles and is provided with fume duct held by a clamp. Temperature ranges from ambient to 350oC controlled by a Sun Vic Energy Regulator.

Micro Kjeldahl Distillation Unit

Weiber micro Kjeldahl distillation double walled unit consists of a heavy casted top made of high grade block of SS304 with room for heating 40 digestion tubes of 75 ml made up of high energy efficient borosilicate glass and this heating is due to heating elements that are made up of high grade kanthal A 1 non magnetic wire, the painted and rust proofed outer casing of this unit is consist of PCRC sheet, the gap in between the walls filled with mineral wool that ensures maximum thermal efficiency.It has a temperature range of 50oC to 350oC and the temperature is controlled through electronic solid state temperature controller cum indicators or microprocessor based digital temperature controller cum indicator. This unit; suitable for the determination of organic nitrogen hence the protein content by the relation % Protein = 6.25 x % using Kjeldahl method that consists of three steps; digestion, distillation and titration.

Soxhlet Extraction Unit

Weiber Soxhlet extraction unit made up of a solvent reservoir extraction assembly with 6 extraction places with concentric metal rings of various diameters to suit 100 ml flasks to 250 ml flasks and built in steam generator and provided with a stand, a clamp holder, Soxhlet extraction (Condenser) glass and extraction flask of 60 ml.It has the temperature range of 50oC to 350oC and the temperature is controlled through electronic solid state temperature controller cum indicator. These units are suitable for extraction of lipids or the compounds having limited solubility in a solvent.

Rotary Flask Shaker

Weiber rotary flask shaker is a compact bench top model having heavy platform fitted with rubber discs to hold conical flasks from 50 ml to 100 ml capacity and is suitable for various research application where shaking of liquid is required in glass flasks. It is mounted on ball bearing crankshafts to give an orbital circular motion to the flasks containing liquids and the entire assembly is mounted on a rental casing which houses a motor coupled by means of V belt with pitch pulleys.

Incubator Shaker

Weiber incubator shaker is a three walled unit, in which the outer wall is made up of thick SS 304, middle layer is of galvanized iron sheet and the inner wall is made up of high grade SS304 and the space between these walls is filled through glass wool for maximal thermal efficiency. It contains beaded elements of nichrome/kanthal A-1 wire are placed in the ribs at the sides and bottom of the second G.I. wall of the chamber  and heaters are placed between the first and the second wall of the shaker, which avoids the direct contact of heaters from the third wall.It  has the temperature range of 5o C to 70o C and temperature is controlled through microprocessor based programmable electronic PID temperature controller cum indicator with sensitivity of +/- 0.5o C or less and it shaking speed is 250 RPM that is controlled through variable speed controller fitted with digital RPM meter.

Seed Germinator as Tissue Culture Equipment

A seed germinator is Tissue/Cell Culture equipment. Using Seed Germinators, varying atmospheric conditions can be created within the chambers without the need of going outside. Typically, these create an environment in which light, humidity and temperature are controlled to provide optimum conditions for the germination of seeds. They are made of double walled structure having two chambers, one inner and one outer. The main components of a Seed Germinator include:

  • A germinating cylinder
  • A temperate controller

Seed Germinators are designed in such a way that they are able to control the crucial environmental parameters.

Tissue/cell culture is a biological method of asexual propagation where a very small piece of tissue (shoot apex, leaf section, or even an individual cell) is excised (which is known as Explants) and placed in sterile /aseptic culture in a test tube/petri dish/tissue culture container containing a special culture media.

A tissue/cell culture is used for rapid multiplication of an organism. A very specialized laboratory with sterile/aseptic conditions is required for tissue culture. All the procedure involved in tissue culture should be carried out in a aseptic conditions.

Steps involved in Tissue/cell Culture are

  • Explants: excision of the plant parts.Place the explants in a tissue culture container/test tubes/petri dishes.
  • Multiplication: Tissue grows and produce small plants.(Rapid multiplication by transfer of fresh culture).

Powder Containment Booths

Powder containment booths control the hazardous emission of powders, dust and vapor without risk to operator or the environment during powder dispensing or sampling processes. In the powder containment booths, down flow of HEPA filtered air prevents the airborne dust particulates form entering into the operator’s breathing zone and prevents the escape of harmful dust particles into the working area. It removes 90% of all particles as small as 5 micron.

Types On the basis of powder dispensing or sampling processes the powder containment booths are of two types:

1.    Downflow Sampling Booth: controls the hazardous emission of powder during powder sampling processes and prevents the worker doing sampling processes from the contaminants. The material that is going to be sampled may belongs to following categories

  • Pharmaceutical products (that can be initial, intermediate and final)

Major applications of Laminar Air Flow

Laminar Air Flow provides a work area with aseptic/sterile conditions. Laminar Air Flow has continuous displacement of air (it provides streamline flow of air) that passes through HEPA (High Efficiency Particulate Air) filter that removes the particulates from the air. Laminar Air Flow are equipped with a UV lamp that should be turned on about 10-20 minutes  before being used to sterilize the shell or cabinet or the surface of the Laminar Air Flow to avoid any kind of contamination. Wipe down the surface with ethanol before and after each use.

Tissue Culture: Laminar Air Flow provides a work area with aseptic/sterile conditions for the tissue culture. Laminar Air Flow has continuous displacement of air (it provides streamline flow of air) that passes through HEPA (High Efficiency Particulate Air) filter that removes the particulates from the air.

In hospitals: Laminar Air Flow is designed for handling of surgical/laboratory materials in a sterile working environment. Sterility in a surgery room is an absolute imperative for ensuring that patients are protected from life-threatening infections. Laminar Air Flow provides a high degree of localized control for critical processes and infections with HEPA filtration. Majority of infections are caused by contact contamination from the patients themselves or the surgical team, studies have also indicated that there is a “relationship between incidence of infection and the level of air contamination”.

Food Processing industries: Main aim of all the food industries is to insure the safety of food that involves the careful control of process from the farm gate to the consumer. Safety includes control of both chemical and microbiological characteristics of the products that is provided by the laminar air flow. Most processing places emphasis on microbial control and often has its objective, the elimination of organisms or prevention of their growth.

Genetic Transformations: Genetic transformation literally means change caused by genes, and involves the insertion of a gene (donor) into an organism (from recipient) in order to change the organism’s trait.  All culture vessels, media and instruments used in protocol must be sterilized. The importance is to keep the air surface and floor free of suspended particles and any other microorganisms. All operations are carried out in Laminar Air Flow, a sterile cabinet.
